Thryv Therapeutics licenses new class of SGK1 inhibitors, expanding into neurodegenerative disease

Thryv Therapeutics Inc, a clinical-stage biopharmaceutical company advancing selective SGK1 inhibitors, announced on Thursday that it has entered into an exclusive, worldwide, royalty-bearing patent and know-how licence agreement with the Spanish National Research Council (Consejo Superior de Investigaciones Científicas, 'CSIC'), the Autonomous University of Madrid (Universidad Autónoma de Madrid, 'UAM') and Fundación de la Universidad Autónoma de Madrid ('FUAM').

This licence adds a structurally distinct SGK1 inhibitor scaffold to Thryv's portfolio, with claims spanning neurodegenerative and cardiovascular indications, creating a purpose-built path toward Parkinson's disease, Alzheimer's disease, and related disorders alongside the company's advancing cardiac programmes.

Thryv holds rights across all fields of use and in all territories, with the right to sublicense.

Thryv's existing clinical SGK1 inhibitor candidates were built through independent medicinal chemistry on a single in-licensed scaffold, supported by a library of more than 500 synthesised analogues and a granted selection patent estate. The CSIC/UAM series is a distinct chemotype arising from a separate discovery programme. Adding a second independent series gives Thryv the ability to match chemistry to indication, continuing to optimise its current molecules for cardiac and cardiometabolic diseases while developing compounds designed from the outset to reach the brain.

The company's lead asset, THRV-1268, is an orally available, selective SGK1 inhibitor currently being evaluated in the WAVE II study in patients with Long QT Syndrome Type 2, with ASPIRE-HF, a Phase 2a study in heart failure with reduced ejection fraction, planned to follow. The newly licensed series is expected to be developed in parallel, with initial characterisation work beginning immediately and a first development candidate targeted for nomination in 2027.
